چکیده

The effect of chemical modification of histidyl residues using diethyl pyrocarbonate (DEP) on plant mitochondrial electron transport was studied. Mitochondrial membranes from potato tubers were isolated and electron flow from NADH to oxygen, NADH to ferricyanide and ascorbate to oxygen were monitored in pro», nee and/or absence of DEP. Measurements were made at various concentrations of DEP and at different pHs either by using an oxygen electrode or spectrophotometrically. The results show that DEP inhibits flow of electrons from NADH to oxygen, however partial electron transport from NADH to ferricyanide and ascorbate to oxygen was unaffected. Maximum inhibition was observed at pH 6.5. The time course of the DEP action revealed a biphasic nature of inhibition. Effects on the levels of reduction of cytochromes b and c by DEP during electron transport indicated that histidyl residues may be present before or at cytochrome b , which are being modified.

Multiple Energy Conservation Bypasses in Oxidative Phosphorylation of Plant Mitochondria

The electron transport chain (ETC) in the inner mitochondrial membrane of most eukaryotes consists of four large protein complexes, the rotenone-sensitive NADH dehydrogenase (complex I), succinate dehydrogenase (complex II), cytochrome bc1 complex (complex III) and cytochrome c oxidase (complex IV). The electron transport activity of complexes I, III, and IV (the latter two often jointly called...

Possible role of free oxidation processes in the regulation of reactive oxygen species production in plant mitochondria.

The non-coupled substrate oxidation mediated by components of the electron transport chain that are not coupled to energy accumulation (such as plant alternative oxidase and rotenone-insensitive NADH dehydrogenases) and uncoupled respiration are peculiar features of plant mitochondria. The physiological significance of such energy-wasting oxidation processes is still debated. It is proposed tha...

Effects of Helminthosporium maydis Race T Toxin on Electron Transport in Susceptible Corn Mitochondria and Prevention of Toxin Actions by Dicyclohexylcarbodiimide.

The effect of Helminthosporium maydis race T toxin on electron transport in susceptible cytoplasmic male-sterile Texas corn (Zea mays L.) mitochondria was investigated, using dichlorophenol indophenol and ferricyanide as electron acceptors. Succinate-dependent electron transport was stimulated by the toxin, consistent with the well described increase in membrane permeability induced by the toxi...

Isolation and oxidative properties of intact mitochondria isolated from spinach leaves.

A procedure was described for preparing intact mitochondria from spinach (Spinacia oleracea L.) leaves. These mitochondria oxidized succinate, malate, pyruvate, alpha-ketoglutarate, and NADH with good respiratory control and ADP/O ratios comparable to those observed with mitochondria from other plant tissues. Glycine was oxidized by the preparations. This oxidation linked to the mitochondrial e...

Plant Mitochondrial Inner Membrane Protein Insertion

During the biogenesis of the mitochondrial inner membrane, most nuclear-encoded inner membrane proteins are laterally released into the membrane by the TIM23 and the TIM22 machinery during their import into mitochondria. A subset of nuclear-encoded mitochondrial inner membrane proteins and all the mitochondrial-encoded inner membrane proteins use the Oxa machinery-which is evolutionarily conser...
